Africa holds more human genetic variation than the rest of the world combined. That fact carries enormous weight for medicine and for understanding our species' past — and far less weight for questions of identity than ancestry marketing implies. It also sits at the centre of an argument about who owns African biological data.

Human populations have lived in Africa longest, so African lineages have had the most time to accumulate mutations, and only a subset of that variation ever left the continent. The consequence is counterintuitive but well established: two people from different African populations can differ from each other more than a European differs from an East Asian. The deepest branches of the human family tree are all inside Africa, and every non-African population is, genetically speaking, a twig on one of them. The variation is also structured by African history. Khoe and San lineages diverged earliest. The Bantu expansion spread a broad West-Central African ancestry across the subcontinent while mixing with everyone it met. Eurasian back-migration shaped North and East African genomes. Indian Ocean trade left South Asian ancestry on the Swahili coast. Madagascar carries an Austronesian component from across an ocean.

African populations carry some of the clearest examples of recent human adaptation. The sickle cell variant, which causes severe disease when inherited from both parents, protects against severe malaria when inherited from one — a brutal evolutionary trade-off that spread precisely where malaria pressure was highest, and which now travels with the diaspora into countries whose health systems are poorly set up for it. Lactase persistence, the ability to digest milk in adulthood, evolved separately in East African herding populations from the well-known European variant: different mutations, same solution, arrived at independently under the same pressure from cattle-keeping. Other examples include variants affecting resistance to trypanosomiasis, adaptation to high altitude in Ethiopian highlanders — achieved by a different physiological route from Tibetan or Andean populations — and immune variation shaped by an exceptionally long history with local pathogens.

For most of the genomic era, African genomes were badly under-represented in research: at various points under two per cent of participants in genome-wide association studies, against a continent holding around eighteen per cent of the world's people and most of its genetic variation. The consequences are practical and harmful. Polygenic risk scores developed on European cohorts transfer poorly to African patients. Pharmacogenomic dosing guidance — for warfarin, for some HIV and tuberculosis drugs, for certain cancer therapies — can be wrong when applied to genotypes the studies never sampled. Variants of unknown significance turn up disproportionately in African patients simply because the reference databases are thin, producing anxiety and misdiagnosis. It is also a missed scientific opportunity. Because African populations carry more variation and shorter blocks of linked DNA, they offer sharper resolution for locating the actual causal variant behind a disease association. Studying African genomes is not only fair; it is methodologically better. H3Africa, funded from 2012 and running projects across more than thirty countries, was built to change this — with a deliberate emphasis on African-led research, African biobanks, African data governance and training African scientists rather than shipping samples abroad.

An ancestry percentage is not a reading of your essence. It is a statistical comparison of your genotype against a reference panel of people whose ancestry the company has assigned by other means. Change the panel and the percentages change. Most companies' African panels are thin and coarse, often lumping vast regions into a handful of categories, which is why results for people of African descent are frequently vague, unstable between companies, and updated dramatically when a panel is revised. The categories themselves need scrutiny. 'Nigerian' is not a genetic group; Nigeria is a state whose borders were drawn by British administrators and amalgamated in 1914, containing hundreds of peoples with different histories. Ethnicity is social, historical and linguistic. Genetics can indicate broad regional ancestry and can sometimes identify relatives, which for descendants of enslaved people can be genuinely meaningful. It cannot tell you your tribe, your culture or who you are.

Race is not a biological unit

Human variation is clinal — it changes gradually across geography — and overlapping, with far more variation inside any named group than between groups. There is no genetic boundary where one race stops and another starts, because the boundaries were drawn by law and custom, not by biology. Skin colour, the trait around which racial classification was built, is controlled by a modest number of genes under strong selection from ultraviolet exposure, and it varies enormously within Africa itself. It carries almost no information about the rest of the genome. This is not a claim that biology is irrelevant to health. Populations differ in the frequency of specific variants, and that matters clinically. The point is that those frequencies do not cluster into races, and using race as a proxy for genotype in medicine — as has been done in kidney function equations and lung capacity measurements — produces documented harm.

This is now the sharpest issue in the field, and it has a history behind it. Samples have repeatedly been collected in Africa under weak consent and used for purposes the donors never agreed to. In 2019 the Wellcome Sanger Institute faced allegations concerning the intended commercial use of African DNA samples without proper consent. The San communities of southern Africa, among the most-studied populations on Earth, responded to a pattern of extractive research by issuing their own research code in 2017, requiring prior community approval, respect and benefit-sharing. The response has been the building of African infrastructure: H3Africa's biorepositories in Nigeria, Uganda and South Africa; national genomics initiatives in Nigeria, Kenya, South Africa, Egypt and Rwanda; data access committees run from the continent; and a growing insistence that African samples stay under African governance and that African scientists appear as senior authors rather than sample providers. The underlying question is the same one that runs through mining and crops in this library: the raw material is African, and the question is where the value and the control end up.

What genetics cannot settle

Genetics is regularly recruited to arguments it cannot decide. It cannot adjudicate claims about which group is indigenous to a territory in any politically useful way, because everybody's ancestors moved and mixed. It cannot validate or invalidate a cultural identity. It cannot rank populations on any trait of social interest, and attempts to do so — in intelligence research especially — have consistently collapsed under scrutiny of their assumptions about heritability and population structure. It also cannot recover most of what was lost in the Atlantic slave trade. A test may indicate broad West African ancestry and occasionally identify living relatives, which is not nothing. It will not name a village, a language or a family, because the records that would connect a genotype to a specific community were mostly never made.

Two things at once. Africa's genomic diversity is a scientific resource of global importance — the reference against which all human variation is measured, and the best available material for finding the biological causes of disease. And the history of how that resource has been sampled, exported and published is a case study in extraction. Getting the second right is what will determine whether the first benefits Africans. A drug developed from African variation should be affordable in the country the variation came from. That is not a sentimental demand; it is what benefit-sharing means in practice, and it is written into instruments like the Nagoya Protocol that remain unevenly enforced.

The continent holds the deepest fossil record of our lineage, the oldest stone tools, the earliest evidence of fire, pigment and ornament, and by far the greatest human genetic diversity on Earth. This is not a claim about African pride; it is the settled reading of the fossil, archaeological and genomic evidence, and it reframes every other chapter that follows.
